Renal Injury Portrait Image Long Description

Overview

Figure 7 illustrates glomerular injury in mouse kidney tissue using both quantitative and qualitative methods. The figure compares different genetic backgrounds for three genes – ASC2, Ifnlrc, and TrpC6 – focusing on how combined mutations affect glomerular structure and injury. It presents a bar graph for injury scoring and representative histological images that highlight morphological differences between genotypes.

Presentation

Three panels are shown:

Panel A (Top left)

A bar graph with the title "Glomerular Injury Score" is plotted on the y-axis, with "Genotype" on the x-axis, subdivided into four groups by the genotype of ASC2, Ifnlrc, and TrpC6 (noted as +/+, +/m, m/m, -/- for each gene). Each bar represents one group, with black dots showing individual sample values. The two groups with m/m and -/- genotypes have significantly higher injury scores compared to the +/+, +/m groups. The graph notes a p-value of <0.0001 by one-way ANOVA, showing strong statistical significance between groups.

Panel B (Top right)

A 40x magnification histological micrograph of a kidney glomerulus from a mouse with ASC2+/+, Ifnlrc+/+, TrpC6+/+ genotype. The tissue is stained, displaying a relatively normal and intact glomerular structure, with clear outlining of the glomerular tuft and Bowman’s space. The texture appears organized and healthy.

Panel C (Bottom row, two images)

Two histological images at high magnification (scale bar = 50 μm), depicting kidney glomeruli from mice with ASC2-/-, Ifnlrc-/-, TrpC6-/- genotype.

  • The left image shows a glomerulus with pathologically widened Bowman’s space, expansion of the mesangial area, and disrupted capillary loops, indicating severe injury and matrix deposition.
  • The right image shows additional glomeruli from the same genotype group, highlighting similar severe architectural disruption, with increased extracellular material and glomerular sclerosis. These findings contrast with the relatively healthy appearance in Panel B.

The figure as a whole demonstrates that mice with triple knockout genotypes experience significantly greater glomerular injury, supported by both quantitative scoring and qualitative histopathological evidence.
